Demineralized bone matrix and Tri-Calcium Phosphate loaded DBM hydrogel for bone regeneration |

Decellularized-demineralized bone matrix (DBM), which contains a wide array of growth factors and cytokines, has been used as a bioactive matrix to promote bone regeneration. However, it requires ancillary material for the extensive use of clinical applications. Herein, we presented demineralized bone matrix particles (DBM) and tri-calcium phosphate powder (TCP) loaded DBM hydrogel (hDBM) scaffold for bone regeneration. Microstructures and morphologies of hDBM, DBM loaded hDBM (hDBM/DBM) and DBM and TCP loaded hDBM (hDBM/DBM/TCP) scaffolds were characterized by SEM. In-vitro studies showed that hDBM/DBM/TCP scaffold was biocompatible and could promote osteogenesis by up-regulating the expression of COL-1, OPN, and Runx2 bone-related genes. But unexpectedly, in-vivo studies using the rabbit-femur defect model revealed that the implanted hDBM/DBM/TCP scaffold was shown a similar pattern to fibrous dysplasia while DBM and TCP granule loaded hDBM scaffold (hDBM/DBM/TCPg) showed good bone formation. Loading TCP granules into DBM hydrogel instead of TCP powder was an effective way of modification of DBM hydrogel promoting bone regeneration. |
