제목 Stimuli-responsive multistage nanoparticles for deep tumor penetration of doxorubicin
초록 Heterogeneous drug distribution within tumor tissue is one of the major hurdles in improving overall therapeutic efficacy. To resolve this issue, we have developed stimuli-responsive multistage nanoparticles, named as transformable nanoparticles (TNPs), composed of pH-sensitive amphiphilic block copolymer, doxorubicin (DOX), and DOX-conjugated ultrasmall gold nanoparticles (DOX-NS). After systemic administration of DOX-loaded TNPs (DOX-TNPs), they were accumulated into the tumor interstitium followed by disassembly of polymeric nanoparticles responding to the mildly acidic tumor environment which triggers the release of DOX and DOX-NS. The released DOX was accumulated in the perivascular region and the DOX-NS could penetrate deeply into the inside of the tumor. After internalization of DOX-NS, DOX would be released by the cleavage of the ester bond. In this study, we successfully demonstrated a great potential of the multistage system of DOX-TNPs in anticancer drug delivery.
