제목 Ultrasound-triggered Drug Delivery Using Glycol Chitosan-based Nanobubble
초록 An ultrasound-based cancer treatment has a great potential for targeted therapy because of its low side effect and higher compatibility. However, the accurate positions to be treated should be required. In this study, we develop gas precursor-loaded polymeric nanobubbles (GCNBs) for ultrasound-triggered drug release with an assistance of contrast-enhanced ultrasound imaging. The nanobubbles were made by hydrophobically-modified glycol chitosan, a shell material, and ultrasound imaging modality and anticancer efficacy were added to encapsulate perfluoropentane (PFP) and docetaxel (DTX) in nanoparticles. The GCNB had an average size around 400nm with the spherical shape, bearing DTX and PFP in inner cores of bubbles. Furthermore, the GCNB showed tumor-homing behavior in tumor-bearing mice model through enhanced permeation and retention (EPR) effect. Also, the treatment using GCNB with ultrasound irradiation achieved a higher therapeutic efficacy on antitumor growth in vivo.
