Effective Bone Regeneration by Periosteum-Derived Osteoblast/BMP-2-Embeded Alginate/Hyaluronic Acid Hydrogel: In Vivo Mini-Pig Model |

Large bone defects are a major challenge for reconstructive surgeons. The use of biological bone grafts is considered as a common therapeutic treatment for the bone regeneration, however, limited availability of donor materials, additional painful procedure for the patients and risk of infection (autografts), in addition to the risk of immune responses and disease transmission (allografts) remain as significant drawbacks in the clinical field. In this study, we prepared injectable human periosteum-derived osteoblast (PO) and/or bone morphogenetic protein-2 (BMP-2)-embedded alginate/hyaluronic acid hydrogels which can allow controlled release of the BMP-2 for the effective bone regeneration. The BMP-2 was continuously released from the hydrogels for more than 35 days. From the in vivo results obtained in a miniature pig model, both PO and BMP-2-embedded alginate/hyaluronic acid hydrogels demonstrated the most effective bone regeneration. |
