Decreased cytotoxcity of PEI/pDNA polyplex with anionic polymer for gene delivery |
초록 |
Among non-viral vectors, polyethylenimine (PEI) is a popular cationic polymer due to high transfection efficiency. Poly (glutamic acid) (PGA) is a hydrophilic and anionic polymer with nontoxicity, biocompatibility, and nonimmunogenicity. To reduce high cytotoxicity of PEI, pDNA/PEI complex was surrounded with PGA. The size of ternary nanoparticle (PEI-pDNA-PGA) was measured by DLS. Its binding ability was determined by gel electrophoresis. In addition, gene transfection efficiency was evaluated by FACS, microscopy, and luciferase assay, resulted in effective vector on gene delivery system.
