The Effect of Bone Differentiation of Chondrocyte in PLGA/Hydroxyaptite/Silk Scaffolds |

Bone is a highly complex tissue which provides mechanical support, and acts as mineral reservoir. We used Poly(lactic-co-glycolic acid)(PLGA) with good biocompatibility and the silk as a natural biomaterial. However, as silk is not an osteogenic material, hydroxyapatite (HAp) with osteoconductive property was added. In the study, PLGA scaffold as a control group, 10 wt% HAp/PLGA, 10 wt% Silk/PLGA, 5 wt% HAp/5 wt% Silk /PLGA (5 wt% H/S/P) and 10 wt% HAp/10 wt% Silk/PLGA (10 wt% H/S/P) scaffolds as experiment groups were prepared. We executed the MTT assay and SEM observation. Also, the degree of differentiation of bone by ALP assay and RT-PCR were analyzed. The analysis of ALP and the result of RT-PCR showed that 10 wt% H/S/P scaffold was significantly increased over time. These results suggest that 10 wt% H/S/P scaffold should serve as structural bases for bone tissue engineering.
