We developed a novel immunoactive nanoparticle based on a metal-phenolic network (MPN) and combined it with electrical ablation to create an effective cancer immunotherapy. MPN nanoparticles were successfully internalized into macrophages, a type of APC, and induced M1 polarization to promote proinflammatory cytokine release. Notably, the novel nanoparticles retained in the tumor region for a longer period of time than the free immune adjuvant. Bipolar electrical ablation tailored to the tumor microenvironment increased therapeutic efficacy by significantly broadening the ablation zone, which further increased immunogenic cell death (ICD). Finally, concurrent treatment with MPN nanoparticles and electrical ablation successfully inhibited tumor growth and prolonged survival in mouse tumor models. This combinational therapy would represent a novel approach to the treatment of intractable cancers. |
