Peripheral artery disease (PAD) is one of the life-threatening vascular diseases that characterized by vascular obstruction, causing ischemia in the hindlimb and reducing blood flow and oxygen supply to the tissues. Ischemic tissues are under oxidative stress associated with significantly high level of reactive oxygen species (ROS) including hydrogen peroxide (H2O2). In this work, we developed H2O2-responsive curcumin-loaded PVAX (CUR-PVAX) nanoparticles which could release bioactive curcumin, vanillyl alcohol and CO2 bubbles in a H2O2-triggered manner. In vivo studies, CUR-PVAX nanoparticles exhibited potent therapeutic actions by suppressing oxidative stress and pro-inflammatory cytokines and enhancing angiogenic factors such as VEGF. In addition, CUR-PVAX nanoparticles displayed enhanced ultrasound contrast by generation of CO2. Herein, we anticipate that CUR-PVAX nanoparticles hold a great translational potential as theranostic agents for ischemic diseases such as PAD. |
