Small interfering RNA (siRNA) is a double-stranded RNA which having 19-25 nucleotides in length. The siRNA is capable of cleavage a target mRNA in mammal so that it can be utilized for target-specific gene silencing mediated gene therapy. However the development of efficient siRNA therapeutics has been impeded due to the unique properties of siRNA such as low charge density and rigidity. To achieve successful gene silencing efficacy, therefore, distinctive and optimized siRNA delivery systems are required. Here, we designed a novel siRNA-crosslinked polymeric nanogel as a siRNA delivery carrier. To release siRNA on intracellular region specifically, we introduced reducible disulfide bond between siRNA and polymers. The synthesis and composition of polymer-siRNA conjugates were analyzed by PAGE method. Also the formation and dissociation of nanogels were confirmed by TEM and SEM images. |
