Naringenin is one of the high value-added compounds in that it is a key precursor for diverse flavonoids and has pharmaceutical activities by itself. For efficient production of naringenin, optimization of multiple gene expressions on the biosynthetic pathways is required, but it is difficult to find optimal balanced expressions. In this context, an integrative way to cover large and balanced expression variations at both transcriptional and translational levels is devised. As a further step, the optimized expression was achieved through naringenin riboswitch-guided high-throughput screening. Through the systematic strategy, a mutant with naringenin productivity 3 times higher than the control strain. It showed 260.2 mg/L naringenin titer, which is the highest value reported so far among glycerol-based production in Escherichia coli. We demonstrate that this multi-level rebalancing based on the biosensors is one of efficient and powerful strategies for pathway optimization. |
