Despite high silencing efficiency and on-target specificity, the clinical translation of siRNA therapeutics has been hindered by its inherent features: poor intracellular delivery, limited blood stability, unpredictable immune responses and unwanted off-targeting effects. For overcoming these hindrances, there have been various efforts to make advances in siRNA itself and its delivery strategies. In our group, we suggested polymerized long-chain siRNA, which can produce highly condensed and homogenous nanocomplexes with different natural polymers, as a new concept for local and systemic siRNA delivery. In this lecture, I will briefly review the history of long-chain siRNA-based delivery systems. Then I will discuss applications of siRNA therapeutics in treatment of different cancers. |
