Gene therapy is defined as insertion of therapeutic genes into cells and tissues to treat different inherited or aquired diseases. Different polymers and synthetic lipids have already been studied as tools for non viral gene therapy. Polyamidoamine (PAMAM) dendrimer is commonly used for gene delivery for its efficient surface modification and DNA condensation property. Although, it is biodegradable and biocompatible, it has shown comparatively lower gene thransfection efficiency than polyethyleneimine (PEI 25KDa). In this study, To enhance the gene expression, we introduced cell penetrating peptides (CPPs) on the surface of PAMAM dendrimer. We synthesized PAMAM G4-histidine-cysteine-arginine (G4-H-C-R) and examined transfection effiency and cytotoxicity of this polymer in cell lines such as HeLa and HEK293. |
