제목 Synthesis of chitosan hydrogel sequentially degrading from the surface by inhibiting penetration of lysozyme via controlling the size and structure of the pores
초록 The drawback in current drug delivery systems is the fact that they control drug releasing rate by simple diffusion. Drug releasing rate by diffusion is not constant over time and such property contributes to side effects when applied to human body. In this study, we designed sequentially and enzymatically degrading chitosan hydrogel for uniform drug releasing rate. After coupled with maleimide group and thiol group, chitosan chains were crosslinked via step reaction to form chitosan hydrogel network. This hydrogel was immersed in Lysozyme solution (pH 7.3) and observed with confocal microscope every single day. We found out that chitosan hydrogel was regularly degraded for two weeks and completely degraded without remaining. Furthermore, time-irrelevantly constant intensity in confocal images of inner part of chitosan hydrogel showed that degradability of the lysozyme was limited inside chitosan hydrogel.
