Among various kinds of proteins, glycan binding proteins (GBPs) play significant roles in many signaling pathways. In spite of their importance, biological properties of GBPs have not been studied well. To study their properties, it is necessary to screen the specific glycan substrate of target GBPs. However, studies for selective separation of GBPs have not been tried. For this purpose, we synthesized a GBP fishing molecule which has three functional groups; aryl hydrazine (for glycan conjugation), diaziridine (for GBPs binding via photoconjugation), and biotin (for separation). With this fishing molecule, we tried to screen the GBPs which were bound to typical glycans. |
