Mussel-inspired immobilization of multiple bioactive molecules for enhanced endothelialization of vascular graft materials |

In this study, we introduced a simple immobilization method of dual bioactive factors, inspired by polydopamine-coating, for the preparation of functionalized vascular graft materials. Polydopamine was deposited on elastic and biodegradable poly(lactic acid-co-ɛ-caprolactone) (PLCL) films, and a cell adhesive RGD-containing peptide and basic fibroblast growth factor(bFGF) were subsequently immobilized by simple dipping. We performed XPS, fluorescamine assay and ELISA for surface characterization. Through fluorescent staining and cell counting, we found that the immobilized molecules had synergistic effects on HUVEC adhesion and proliferation. In Western blot and RT-PCR analysis, promoted expression of endothelialization markers was observed that was influenced by co-immobilized factors. In conclusion, the polydopamine layer is effective to immobilize multi-bioactive molecules as an active form and co-immobilized molecules can have synergistic effects on HUVEC behaviours. |
